Background: Diabetes mellitus is a metabolic disorder characterized by hyperglycemia. Delays in blood sample processing can induce glycolysis, leading to decreased serum glucose concentration; thus, the choice of blood collection tube is critical. Objective: To determine the difference in serum glucose levels between Clot Activator Tubes and Serum Separator Tubes (SST) after a 6-hour storage delay. Methods: This quasi-experimental study was conducted at RS Bunda Palembang from June 12 to 19, 2025. A total of 42 specimens were selected using purposive sampling. Glucose levels were evaluated immediately and compared with those measured after a 6-hour delay across both tube types. Results: The mean glucose level tested immediately was 202.04 mg/dL. Following a 6-hour delay, the mean glucose level dropped to 182.50 mg/dL in Clot Activator Tubes and 190.16 mg/dL in SSTs. Statistical analysis revealed a significant difference in glucose stability between the two tube types (p < 0.001). Conclusion: There is a significant difference in serum glucose levels between Clot Activator Tubes and Serum Separator Tubes after a 6-hour delay, with SSTs demonstrating superior performance in minimizing glucose degradation.
