Abstract

Microscopic examination of fecal specimens using the flotation method has higher sensitivity than direct smear and sedimentation methods. Centrifugal flotation enables the detection of a greater number of helminth eggs and reduces the likelihood of false-negative results by facilitating the rapid sedimentation of interfering particles. The use of a saturated sucrose solution also provides a cleaner and clearer microscopic background. This study was conducted using fecal specimens containing Ascaris lumbricoides eggs to determine the optimal flotation time for recovering the highest number of eggs. Three positive fecal specimens were examined using centrifugal flotation with flotation times of 20, 30, and 40 minutes, each performed in five replicates. The recovered flotation layer was examined microscopically, and the number of Ascaris lumbricoides eggs was counted and averaged. Data were analyzed using the Shapiro–Wilk normality test followed by a one-way analysis of variance (One-Way ANOVA). The mean egg counts were 118 eggs at 20 minutes, 162 eggs at 30 minutes, and 134 eggs at 40 minutes of flotation. The One-Way ANOVA revealed a statistically significant difference among the flotation times (p = 0.000, α = 0.05). Variations in flotation time using the centrifugal flotation method significantly affected the number of Ascaris lumbricoides eggs recovered. Therefore, laboratory personnel are recommended to use a flotation time of 30 minutes with saturated sucrose solution when performing the centrifugal flotation method to achieve optimal egg recovery.
